CaCl₂ is used to prepare bacterial cells for transformation by inducing competence. The calcium ions neutralize negative charges on the bacterial cell membrane and DNA, facilitating the binding of DNA to the cell surface. This treatment also disrupts the membrane structure, making it more permeable to DNA. Combined with heat shock, this process enables the DNA to enter the bacterial cells efficiently.
