To collect rhizosphere soil of seagrasses, gently uproot plants and shake off loosely attached sediment. The remaining sediment closely adhering to roots is considered rhizospheric. It’s then brushed, washed, or sonicated into sterile containers. The process must be done quickly in field or lab to prevent microbial shifts. Salinity and anoxic conditions must be preserved to reflect natural habitat conditions.
