1. Cartilage
- Light Microscopy:

- In hyaline cartilage, there may be a matrix with fine collagen fibers, appearing more homogenous and translucent.
- Electron Microscopy:
- Cartilage will show well-defined chondrocytes and a dense extracellular matrix with a fibrous appearance due to collagen fibers.
2. Ciliated Epithelium
- Light Microscopy:
- Look for a layer of epithelial cells that appear columnar or cuboidal. The cilia will be visible as tiny hair-like projections on the apical surface.
- The cells will have a nucleus located near the base, and the entire layer will appear organized.
- Electron Microscopy:
- You will see cilia as prominent, elongated structures at the surface of the epithelial cells, and the organization of microtubules (9+2 arrangement) within the cilia can be visualized.
3. Goblet Cells
- Light Microscopy:
- Goblet cells can be identified as single, mucous-secreting cells interspersed among the ciliated epithelial cells. They appear as oval or flask-shaped structures that may look clear or slightly stained (depending on the staining method) due to their mucus content.
- Electron Microscopy:
- Goblet cells will show a well-defined apical region containing mucus granules and a nucleus located toward the base of the cell.
4. Squamous Epithelium of Alveoli
- Light Microscopy:
- Look for a thin layer of flat cells (pavement-like appearance) that line the alveoli. The cells will be much thinner than cuboidal or columnar epithelial cells and may appear as a single layer.
- Electron Microscopy:
- The squamous epithelium will show very thin cytoplasmic extensions, with minimal organelles, allowing for a high surface area for gas exchange. You may also see adjacent alveoli and the close association with capillaries.
5. Smooth Muscle
- Light Microscopy:
- Smooth muscle fibers will appear as elongated, spindle-shaped cells with centrally located nuclei. The cells may be arranged in bundles or layers.
- The staining may reveal a faint, wispy appearance, often paler than striated muscle.
- Electron Microscopy:
- You will see a more homogeneous appearance with elongated cells, less organelle density compared to striated muscle, and prominent cytoplasmic filaments.
6. Capillaries
- Light Microscopy:
- Capillaries may not be distinctly visible on their own, but you can identify them by observing clusters of red blood cells within small, thin-walled vessels that are often close to the alveoli or other tissues.
- They appear as tiny, round, or oval structures within tissues, often surrounding the alveolar walls.
- Electron Microscopy:
- Capillaries will show endothelial cells that form a thin layer (one cell thick) surrounding a lumen with red blood cells. The close association with surrounding tissues is important for facilitating gas exchange.
